Human Frontiers Science Programme – “Ribosomal signalling and specificity in co-translational protein processing and folding”

You are invited to join an international, multi-centre, interdisciplinary project funded by the Human Frontiers Science Programme. Our goal is to test the hypothesis that the ribosome plays a central role sensing the nature of the emerging nascent chain within the eukaryotic ribosomal tunnel and also that it is involved in signalling to the exterior of the ribosome to recruit the appropriate nascent polypeptide-associated factors (NAFs).

To test this hypothesis, we will investigate the conformational properties of nascent polypeptides at different stages of elongation in the presence and absence of different NAFs to map their folding properties. Specifically, you will be involved in the development and application of mass spectrometry-based techniques using ion mobility mass spectrometry, chemical modification and H/D exchange monitored by Mass Spectrometry (MS), combined with other biophysical analyses, to address these questions.

This project is between the laboratories of Professors Sheena Radford and Alison Ashcroft within the Astbury Centre for Structural Molecular Biology, in collaboration with Professors Elke Deuerling (University of Konstanz, Germany) and Judith Frydman (Stanford University, USA).

You should have, or be close to completing, a PhD in Chemistry, Biochemistry, Biophysics or a related discipline. Experience in protein mass spectrometry and non-covalent interactions in complex assemblies is essential.
