A 2-year postdoctoral research position is available at the University Lille 2 France’s EGID (European Genomic Institute for Diabetes http://www.egid.fr/) to develop induced Pluripotent Stem (iPS) Cell technology as a disease model for type 2 diabetes. Over the last 10 years, the U859 (F. Pattou) and UMR8199 (P. Froguel) laboratories in Lille have analyzed the effect of genetic type 2 diabetic-related variants on human pancreatic function both in an obesity cohort and more recently via exploitation of a database of research and clinical grade pancreatic islet preparations where archived paraffin sections allowed retrospective genotyping. Clinical cohorts include longitudinal clinical data on diabetes parameters and biological tissue except the pancreas. The iPS cell technology could bridge the gap allowing us to study in vitro and in vivo patient derived- iPS- insulin secreting cells and perform genotype-phenotype studies.
Our main aims are to establish hiPSC disease modeling technology in Lille by 1) building a trained human iPSC team in Lille and acquiring differentiation technology to generate beta cells in vitro and in vivo from hiPSC in collaboration with a US lab 2) To define which part of the variation in the disease model is due to the experimental procedures and which part of the variation may be due to genetic background, We will characterize the properties of the iPS-insulin secreting cells using in vitro and in vivo adaptation assays in comparison to islets.
